Over the last few years there has been substantial discourse on the bodybuilding forums regarding whether or not this particular product is actually a 95% 3,4-divanillyltetrahydrofuran extract, or whether this is simply a highly concentrated ratio extract of nettle root. Barlowe's is making no specific claim regarding the exact breakdown of this Nettle Extract. 

    Each capsule contains 300 milligrams of the 95% high potency nettle root extract and 150 milligrams of the 5%/2% stantardized nettle root extract. The 5%/2% nettle extract (also of very high quality) is primarily added in order to make the encapsulation process proceed more easily, as the 95% nettle in its pure form is difficult to work with. One capsule taken twice a day is the therapeutic dosage needed to optimally bind SHBG and increase free testosterone in someone of normal physical condition.  Although Nettle (Urtica dioica) has been utilized for hundreds of years to treat a variety of ailments, it is primarily of interest now because it has been discovered that it is able to increase free testosterone in the bloodstream, and 3,4-divanillyltetrahydrofuran is the substance in Nettle which primarily causes this effect.

    For many men, a safe and easy way to increase free testosterone is to prevent it from being converted (aromatized) into estrogen. Too much estrogen plays havoc with a man's sex life by binding to testosterone receptor sites and may contribute to the over-production of sex hormone-binding globulin (SHBG). SHBG binds free testosterone in a way that makes it unavailable to receptor sites in the brain, nerves and genitals. High serum levels of estrogen also trick the brain into thinking that enough testosterone is being produced, thereby slowing the natural production of testosterone. Some studies show that the decline in sexual interest with advancing age is not always due to the amount of testosterone produced, but rather to the increased binding of testosterone to globulin by SHBG. This explains why some older men who are on testosterone replacement therapy do not report a long-term aphrodisiac effect. The artificially administered testosterone becomes bound by SHBG, and is not available to cellular receptor sites where it would normally produce a libido-enhancing effect.

    Logically, one way of increasing libido in older men would be to block the testosterone-binding effects of SHBG. This would leave more testosterone in its free, sexually activating form. A highly concentrated extract from the Nettle root provides a unique mechanism for increasing levels of free testosterone. Recent European research has identified constituents of Nettle root that bind to SHBG in place of testosterone, thus reducing SHBG's binding of free testosterone. Therefore, this product is an excellent herbal extract to take in combination with Barlowe's Tongkat Ali, Bulbine Natalensis, or Fenugreek Extracts, as the Tongkat, the Bulbine, and the Fenugreek dramatically increase level of testosterone, and the High Potency Nettle Extract keeps the raised testosterone in its free, powerful form.

    Lignans bind to sex hormone-binding globulin (SHBG). The lignan with the highest binding affinity is (+/ )-3,4-divanillyltetrahydrofuran. In a double Stobbe condensation--without use of protecting groups--a wide variety of lignans with different substitution pattern in the aromatic and aliphatic part of the molecule was synthesized. These lignans were tested in a SHBG-binding assay which allowed to deduce the following relationship between structure and activity: 1) (+/-)-diastereoisomers are more active than meso compounds 2.) the 4-hydroxy-3-methoxy (guajacyl) substitution pattern in the aromatic part is most effective 3.) the activity increases with the decline in polarity of the aliphatic part of the molecule.

    Polar extracts of the stinging nettle (Urtica dioica L.) roots contain the ligans (+)-neoolivil, (-) secoisolariciresinol, dehydrodiconiferyl alcohol, isolariciresinol, pinoresinol, and 3,4 divanillyltetrahydrofuran. These compounds were either isolated from Urtica roots, or obtained semisynthetically. Their affinity to human sex hormone binding globulin (SHBG) was tested in an in vitro assay. In addition, the main intestinal transformation products of plant lignans in humans, enterodiol and enterolactone, together with enterofuran were checked for their activity. All lignans except (-) pinoresinol developed a binding affinity to SHBG in the in vitro assay. The affinity of (-)-3,4 divanillyltetrahydrofuran was outstandingly high. These findings are discussed with respect to potential beneficial effects of plant lignans on benign prostatic hyperplasia (BPH).
